Layton receives a letter from Jenice Kaitly, a famous opera singer, asking them to witness her latest performance at the beginning of the movie. She also seeks help from Layton, Luke, and Remi in solving a mystery, claiming that her friend Melina vanished a year ago and reappeared as a seven-year-old girl, claiming to have discovered the secret to endless life. Layton and his trainees are naturally enthralled by the situation, so they journey to the Crown Petone opera theatre. When they arrive, they notice something odd: the pianist, Ozro Vislar, mentions a game in which the winner will be granted "eternal life." Before it's too late, Layton, Luke, and Remi must discover the riddle of the Eternal Diva.
